Kher belongs to a middle-class Kashmiri Pandit family.
Anupam was not good at studies and sports in his childhood, he was only good in drama and theatre acting.
He met Kirron Kher in a theatre at Chandigarh, they become best friends later.
In Panjab University’s Department of Theatre, he performed Balwant Gargi and Amal Allana theatre play, for which he got a gold medal, and also got admission in National School of Drama.
His 1984 film Saaransh was the first milestone for him in his acting career, in which he performed an old-man role. After this success of Saaransh, he was offered around 100 films in just two weeks.
In the Raj Se Swaraj Tak television series, he performed a role as Mahatma Gandhi.

He made a record for being the only one for getting five Filmfare awards for best acting in a Comic Role.
Kher performed the role of Manmohan Singh (former Prime Minister of India) in The Accidental Prime Minister, Anupam said that it was the most difficult role.
